Formulation development and evaluation of controlled release gastro-retentive advance delivery system of furosemide

Abstract
The most important use of furosemide is in the treatment of hypertension or pulmonary edema because no other group of diuretics is more effective than the loop diuretics in this situation. But there are certain major drawbacks with the conventional therapy of furosemide.Optimized formula was selected by desirability (numerical optimization) technique. Three formulation batches of the predicted optimized formulation of gastroretentive bilayer tablet were prepared and experimentally validated by comparision method. Comparison of the observed and software predicted responses showed a good correlation. This demonstrated that the factorial design optimization technique was successful in the formulation development of gastroretentive layer of furosemide. Finally bilayer tablets were prepared using optimized solid dispersion and optimized floating layer. The final bilayer tablet was evaluated and was found to be within the limits.
